چکیده

Ingestion of pure fructose stimulates de novo lipogenesis and gluconeogenesis. This may however not be relevant to typical nutritional situations, where fructose is invariably ingested with glucose. We therefore assessed the metabolic fate of fructose incorporated in a mixed meal without or with glucose in eight healthy volunteers. Each participant was studied over six hours after the ingestion of liquid meals containing either 13C-labelled fructose, unlabeled glucose, lipids and protein (Fr + G) or 13C-labelled fructose, lipids and protein, but without glucose (Fr), or protein and lipids alone (ProLip). After Fr + G, plasma 13C-glucose production accounted for 19.0% ± 1.5% and 13CO2 production for 32.2% ± 1.3% of 13C-fructose carbons. After Fr, 13C-glucose production (26.5% ± 1.4%) and 13CO2 production (36.6% ± 1.9%) were higher (p < 0.05) than with Fr + G. 13C-lactate concentration and very low density lipoprotein VLDL 13C-palmitate concentrations increased to the same extent with Fr + G and Fr, while chylomicron 13C-palmitate tended to increase more with Fr + G. These data indicate that gluconeogenesis, lactic acid production and both intestinal and hepatic de novo lipogenesis contributed to the disposal of fructose carbons ingested together with a mixed meal. Co-ingestion of glucose decreased fructose oxidation and gluconeogenesis and tended to increase 13C-pamitate concentration in gut-derived chylomicrons, but not in hepatic-borne VLDL-triacylglycerol (TG). This trial was approved by clinicaltrial. gov. Identifier is NCT01792089.

Fructose metabolism in humans – what isotopic tracer studies tell us

Fructose consumption and its implications on public health are currently under study. This work reviewed the metabolic fate of dietary fructose based on isotope tracer studies in humans. The mean oxidation rate of dietary fructose was 45.0% ± 10.7 (mean ± SD) in non-exercising subjects within 3-6 hours and 45.8% ± 7.3 in exercising subjects within 2-3 hours. When fructose was ingested together ...

Effect of Fructose Intake on Metabolic Biomarkers in Adults with Type 2 Diabetes Mellitus: A Systematic Review of Randomized Clinical Trial Studies

Introduction: Fructose is a lipogenic monosaccharide affecting glucose homeostasis and other metabolic biomarkers; however, there is conflicting evidence in this regard. The current systematic review aimed to determine the effects of fructose on metabolic biomarkers in individuals with T2DM using randomized clinical trial studies. Materials and Methods: In this systematic review study, after se...

Central and Metabolic Effects of High Fructose Consumption: Evidence from Animal and Human Studies

Fructose consumption has increased dramatically in the last 40 years, and its role in the pathogenesis of the metabolic syndrome has been implicated by many studies. It is most often encountered in the diet as sucrose (glucose and fructose) or high-fructose corn syrup (55% fructose). At high levels, dietary exposure to fructose triggers a series of metabolic changes originating in the liver, le...

A high-fat, high-fructose diet accelerates nutrient absorption and impairs net hepatic glucose uptake in response to a mixed meal in partially pancreatectomized dogs.

The aim of this study was to elucidate the impact of a high-fat, high-fructose diet (HFFD; fat, 52%; fructose, 17%), in the presence of a partial (~65%) pancreatectomy (PPx), on the response of the liver and extrahepatic tissues to an orally administered, liquid mixed meal. Adult male dogs were fed either a nonpurified, canine control diet (CTR; fat, 26%; no fructose; n = 5) or a HFFD (n = 5) f...

Troxerutin Chronic Treatment Protects against Fructose-induced Metabolic Syndrome in Male Rats

Background and Aims: Metabolic syndrome, a common metabolic disorder, for the serious health consequences such as insulin resistance and lipid abnormalities has been considered as a major clinical challenge with obscure causes. Oxidative stress is an important component of metabolic syndrome, contributing in its development. Troxerutin, a semi-synthetic derivative of natural bioflavonoid rutin,...
